1-Change in the English and Arabic titles:
Purslane Leaf Extract: A Novel Therapeutic Approach to Mitigate the Effect of 4 (Methylnitrosamino)-1-(3-pyridyl)-1-buta-4 None (NNK)-Induced on Testicular Histopathological Alterations and Hormonal Imbalances in Male Rats.
مستخلص أوراق البورسلين: نهج علاجي جديد للتخفيف من تأثير 4 (ميثيلنيتروزامينو)-1-(3-بيريديل)-1-بوتا-4 غير مستحثة (NNK) على التغيرات النسيجية المرضية في الخصية واختلالات هرمونية في ذكور الفئران.
2-Duration: 2 months is insufficient to complete the in vivo experiments, blood analysis, and histopathology.
3-The team is complementary and can achieve the work.
4- The fund must be added; they wrote that it will be covered by themselves without informing of the exact expenses.
5-Authors have approval from the department and institute, but they say their research work was not reviewed.
6-Objective is the Investigation of bioactive constituents from the plant for possible medicinal use. For that, my suggestion is that the authors can do HPLC for the extract to detect the active constituent in the plant that affects fertility
7-Methods:
7.1-What is the exact part of the plant that the authors will be extracting? Is it the leaves, stem, or the roots?
7.2- The authors must write the references to confirm the doses of the purslane extract and NNK that they will use.
7. 3- The authors need to assess the cytotoxicity of Purslane extract and NNK doses. They should perform either an LD50 test or an in vitro assay, such as the MTT assay, to determine the half maximal inhibitory concentration (IC50).
7.4-The site of blood collection must be confirmed, whether it is from the retro-orbital venous plexus or the abdominal aorta; the authors mention both methods in the protocol.
7.5-The authors did not address the risks of their work.
